Leptospirosis is an infectious disease caused by the pathogenic bacterial genus Leptospira sp. Leptospirosis can be transmitted by domestic animals such as dogs, cats, pigs, cows and rodents, especially mice. Leptospirosis is transmitted by leptospira bacteria which are widespread throughout the world and are transmitted directly or indirectly from animals to humans (zoonosis). Human-to-human transmission is possible, but contact with environments such as leptospira-contaminated soil and water is very rare. Samples were taken based on the KAP (Knowledge, Attitude and Practice) survey, Knowledge, Attitude, differentiating practices or actions into 3 levels, namely Guided Response, Mechanism, Adoption, samples taken were Officers Cleanliness, dump truck driver, armroll driver, crew, sweeper 319 employees. Sampling was carried out using a simple random sampling method using interviews. Based on the results of the research and discussion, it can be concluded that the knowledge of cleaning staff regarding the dangers of leptospirosis in the Kupang City Environment and Hygiene Service is dominated by knowledge in the sufficient category of 60% (24/40) of respondents. The attitude of cleaning officers at the Kupang City Environment and Hygiene Service is dominated by the fair category at 70% (28/40) of respondents. The practices of cleaning staff at the Kupang City Environment and Hygiene Service are dominated by the adequate category at 90% (36/40) of respondents.
